The purpose of this study was to examine the acute effects of maximal concentric vs. eccentric exercise on the isometric strength of the elbow flexor, as well as the biceps brachii muscle electromyographic (EMG) responses in resistance-trained (RT) vs. untrained (UT) men. Thirteen RT men (age: 24 ± 4 years; height: 180.2 ± 7.7 cm; body weight: 92.2 ± 16.9 kg) and twelve UT men (age: 23 ± 4 years; height: 179.2 ± 5.0 cm; body weight: 81.5 ± 8.6 kg) performed six sets of ten maximal concentric isokinetic (CON) or eccentric isokinetic (ECC) elbow flexion exercise in two separate visits. Before and after the exercise interventions, maximal voluntary contractions (MVCs) were performed for testing isometric strength. In addition, bipolar surface EMG signals were detected from the biceps brachii muscle during the strength testing. Both CON and ECC caused isometric strength to decrease, regardless of the training status. However, ECC caused greater isometric strength decline than CON did for the UT group (p = 0.006), but not for the RT group. Both EMG amplitude and mean frequency significantly decreased and increased, respectively, regardless of the training status and exercise intervention. Resistance-trained men are less susceptible to eccentric exercise-induced muscle damage, but this advantage is not likely linked to the chronic resistance training-induced neural adaptations. 相似文献

Our recent studies implicate the transient receptor potential vanilloid-1 (TRPV1) channel as a mediator of retinal ganglion cell (RGC) function and survival. With elevated pressure in the eye, TRPV1 increases in RGCs, supporting enhanced excitability, while Trpv1 -/- accelerates RGC degeneration in mice. Here we find TRPV1 localized in monkey and human RGCs, similar to rodents. Expression increases in RGCs exposed to acute changes in pressure. In retinal explants, contrary to our animal studies, both Trpv1 -/- and pharmacological antagonism of the channel prevented pressure-induced RGC apoptosis, as did chelation of extracellular Ca2+. Finally, while TRPV1 and TRPV4 co-localize in some RGC bodies and form a protein complex in the retina, expression of their mRNA is inversely related with increasing ocular pressure. We propose that TRPV1 activation by pressure-related insult in the eye initiates changes in expression that contribute to a Ca2+-dependent adaptive response to maintain excitatory signaling in RGCs. 相似文献

ADP-glucose pyrophosphorylase (AGP) represents a key regulatory step in polysaccharide synthesis in organisms ranging from bacteria to plants. Higher plant AGPs are complex in nature and are heterotetramers consisting of two similar but distinct subunits. How the subunits are assembled into enzymatically active polymers is not yet understood. Here, we address this issue by using naturally occurring null mutants of the Shrunken2 (Sh2) and Brittle2 (Bt2) loci of maize as well as the yeast two-hybrid expression system. In the absence of the maize endosperm large AGP subunit (SH2), the BT2 subunit remains as a monomer in the developing endosperm. In contrast, the SH2 protein, in the absence of BT2, is found in a complex of 100 kD. A direct interaction between SH2 and BT2 was proven when they were both expressed in yeast. Several motifs are essential for SH2:BT2 interaction because truncations removing the N or C terminus of either subunit eliminate SH2:BT2 interactions. The physical mechanisms underlying the transport of ions across a model potassium channel are described. The shape of the model channel corresponds closely to that deduced from crystallography. From electrostatic calculations, we show that an ion permeating the channel, in the absence of any residual charges, encounters an insurmountable energy barrier arising from induced surface charges. Carbonyl groups along the selectivity filter, helix dipoles near the oval chamber, and mouth dipoles near the channel entrances together transform the energy barrier into a deep energy well. Two ions are attracted to this well, and their presence in the channel permits ions to diffuse across it under the influence of an electric field. Using Brownian dynamics simulations, we determine the magnitude of currents flowing across the channel under various conditions. The conductance increases with increasing dipole strength and reaches its maximum rapidly; a further increase in dipole strength causes a steady decrease in the channel conductance. The current also decreases systematically when the effective dielectric constant of the channel is lowered. The conductance with the optimal choice of dipoles reproduces the experimental value when the dielectric constant of the channel is assumed to be 60. The current-voltage relationship obtained with symmetrical solutions is linear when the applied potential is less than approximately 100 mV but deviates from Ohm's law at a higher applied potential. The reversal potentials obtained with asymmetrical solutions are in agreement with those predicted by the Nernst equation. The conductance exhibits the saturation property observed experimentally. We discuss the implications of these findings for the transport of ions across the potassium channels and membrane channels in general. 相似文献

The boll weevil (Anthonomus grandis Boheman) is an insect pest of cotton that underwent a well-documented range expansion across the southeastern U.S. from Mexico
beginning about 110 years ago. Eleven microsatellite loci were surveyed to infer the magnitude and pattern of genetic differentiation
among boll weevil populations from 18 locations across eight U.S. states and northeast Mexico. Estimates of genetic diversity
(allelic diversity and heterozygosity) were greater in Southern than Northern populations, and were greater in the west than
the east among Northern populations. Boll weevil populations were genetically structured as a whole across the geographic
range sampled, with a global F
ST of 0.241. South-central populations exhibit classic isolation by distance, but evidence suggests that populations within
the Eastern and Western regions have not yet reached genetic equilibrium. Gene flow appears to be relatively high among populations
within the Eastern region. Population assignment data and estimates of gene flow indicate that migration between locations
separated by < 300 km is frequent. The database of microsatellite genotypes generated in this study now makes it possible,
through population assignment techniques, to identify the most likely geographic source of a boll weevil reintroduced to an
eradication zone, which will help action agencies decide the most appropriate mitigation response.

Pheromone-baited traps are often used in ecological studies of the European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae). However, differences in trap captures may be confounded by trap design, trap location relative to a windbreak, and changes in local weather. The objectives of this experiment were, first, to examine differences in 0. nubilalis adult (moth) captures among the Intercept wing trap, the Intercept bucket/funnel UNI trap, and the Hartstack wire-mesh, 75-cm-diameter cone trap (large metal cone trap) as well as among three cone trap designs. Second, we examined the influence of the location of the large metal cone trap relative to a windbreak on the number of moths captured. Third, we examined the relationship between nightly mean air temperature, relative humidity, wind speed, precipitation, and the number of moths captured in large metal cone traps. The number of moths captured was significantly influenced by trap design, with large metal cone traps capturing the most moths. Wing and bucket traps were ineffective. Differences among trap captures were significant among trap locations relative to a windbreak. Under strong (>14 kph) or moderate (7 <14 kph) wind speeds, traps located leeward of the windbreak captured the most moths, but when wind speeds were light (<7 kph), traps not associated with windbreaks captured the most moths. The multiple regression model fitted to the relationship between number of moths captured per Julian date and nightly weather patterns was significant. Interest in the ecological and population genetics of the western corn rootworm, Diabrotica virgifera virgifera LeConte, has grown rapidly in the last few years in North America and Europe. This interest is a result of a number of converging issues related to the increasing difficulty in managing this pest and the need to characterize and understand gene flow in the context of insect resistance management. One of the key components needed for successful population genetics studies is the availability of suitable molecular markers. Using a standard group of microsatellite markers enables researchers from different laboratories to directly compare and share their data, reducing duplication of effort and facilitating collaborative work among laboratories. We screened 22 candidate microsatellite loci against five criteria to create a core set of microsatellite markers for D. v. virgifera population genetics studies. The criteria for inclusion were moderate to high polymorphism, unambiguous readability and repeatability, no evidence of null alleles, apparent selective neutrality, and no linkage between loci. Based on our results, we recommend six microsatellite markers to be included as a core set in future population genetics studies of D. v. virgifera along with any other microsatellite or genetic markers. As more microsatellites are developed, those meeting the criteria can be added to the core set. We encourage other groups of researchers with common interests in a particular insect species to develop their own core sets of markers for population genetics applications. 相似文献
